Preclinical evidence suggests that simultaneous inhibition of the anandamide-degrading enzyme FAAH and the prostaglandin-generating enzyme cyclooxygenase (COX) produces greater analgesic efficacy and fewer side effects than either mechanism alone. We have combined computational and biophysical methods to discover multi-target FAAH/COX inhibitors with potential application in colon cancer and chronic inflammation. Two distinct classes of FAAH/COX blockers have been identified, which are currently undergoing further chemical and pharmacological investigation.
