The multifaceted nature of Alzheimer’s disease requires innovative approaches to the discovery of safe and effective new drugs. The goal of this project is to identify dual inhibitors of acetylcholinesterase (AChE) and extrasynaptic NMDA receptor channels as candidates for Alzheimer’s therapy. Preclinical and clinical evidence suggests that such compounds might interact synergistically to improve cognition. We have identified a first series of chemical hits in this class and are using both computational methods and classical structure-activity relationship analyses to optimize them into preclinical candidates.
